Abstract:

Background: Epididymal sperm has been recovered with the aim of conserving potentially valuable genetics Materialss from post mortem males for subsequent usage in assisted reproduction technologies. Among the quality parameters of spermatozoa, motility and velocity patterns are sensitive for chemical and physical components of medium which they were diluted. Also close correlation between sperm motion characteristics and sperm fertility has been found. So the aim of this study was to evaluate, motion characteristics of buffalo epididymal spermatozoa preserved in different culture media during 6 hour incubation. Materials and Methods: Thirty testes from 15 mature buffalo bulls (3-5 year) were collected immediately after animal’s death from Urmia slaughterhouse and transported to laboratory in cool condition. Spermatozoa were collected from caudal epididymis and pooled together. The samples were diluted in Ham’s F10, human tubal fluid (HTF), Tissue culture medium (TCM) with 7.2 PH and concentration of 20×106 sperm/ml. sperm motion characteristics were evaluated 1, 3 and 6 hours of incubation at 37 °C with computer assisted sperm analyzer(CASA HFT version 6.0). Results: Result showed that, all of the kinetics parameters reduced dramatically in time depend manner. Preservation of epididymal sperm in HTF and TCM media yielded higher motility and progressive motility as compared with Ham’s F10 at 1, 3 and 6 hours of preservation (p<0.05). Velocity patterns (straight line, curvilinear and average path velocity) were highest in HTF medium and lowest in Ham’sF10 medium in all of the incubation time (p<0.05). Conclusion: It can conclude that preservation of buffalo epididymal spermatozoa in HTF medium at 37°C seems to be suitable for sperm motility and velocity patterns.

P-119: In Vitro Assessment of Sucrose for Preservation of Buffalo Epididymal Spermatozoa

Background: Epididymal spermatozoa have been used for production of offspring from genetically important animals after they are dead. To attain optimal fertility with epididymal spermatozoa, they should be preserved in a suitable medium in 37°C. Preservation media provides requirement for spermatozoa and affects its motility, viability, and fertility. Sucrose is one of the important components ...

P-63: The Effect of Temperatures on Goat Epididymal Sperm Motility in Tissue Culture Media 199

Background: There is a general deterioration in sperm quality depending on time postmortem. Refrigeration of the epididymes down to around 5°C is the best strategy to lower this damage and there are many dissim ilarities between species, possibly due to differences on cold shock endurance of epididymal sperm. Our objective was to determine the effect of 5°C and 37°C on goat epididymal sperms mo...

P-133: Effect of Thawing Rate on Kinetic Parameters of Frozen - Thawed Buffalo Spermatozoa Extended in Tris - Egg Yolk

Background: Optimal thawing rate is critical for quality and fertilizability of frozen -thawed spermatozoa. Kinetic parameters are the most important factors which are affected by thawing rate so, that have close relation between kinetic parameters and fertility of the spermatozoa. Tris - egg yolk have been widely used for cryopreservation of the buffalo and other species semen. According to ou...

P-116: The Catalase Effect on LDH Enzyme Leakage of Buffalo Epididymal Sperm During Culture in Human Tubal Fluid Mediaa

Background: Mammalian spermatozoa are highly susceptible to lipid peroxidation, which leads to structural damage to the sperm cell, the enzymatic leakage from spermatozoa accompanied by lowered motility and metabolism. Catalase, an enzyme that detoxify hydrogen peroxide and reduce the enzyme leakage from sperm cell Materials and Methods: For this study, buffalo bull testicles (20 pairs) were pi...

P-134: In Vitro Assessment of Cysteine Effect on DNA Integrity of Frozen-Thawed Buffalo Spermatozoa

Background: Cysteine has been used as additive in cryopreservation extenders. It contains of thiol group which plays an antioxidant role to eliminate reactive oxygen species (ROS) during freeze -thaw procedure. Excessive ROS production cause DNA damage and subsequently reduce sperm fertility. On the other hand, protamine protein have high levels of cysteine which is critical in packaging and co...

P-70: Effect of Xylose on Epididymal Sperm Motility in Water Buffalo (Bubalus bubalis)

Background: In mammalian spermatozoa, motility is one of the most important parameters for evaluating the quality of the semen. From a functional point of view, motility is strongly related to the spermatozoa’s ability to manage its energy status, Thus, modulation of energy status could be translated into significant changes in motility patterns. The aim of the present study was to evaluate xyl...
